Bio-Adembeads (300nm) coated with recombinant high quality Protein A covalently coupled to the surface. Protein A is a cell wall component composed of a single polypeptide chain of molecular weight 42 kDa produced by Staphylococcous aereus (Cowan Strain). The protein A employed is recombinant form containing the Fc binding sites without any albumin binding sites. Protein A is capable of binding to the Fc portion of immunoglobulins, especially IgGs, from a large number of species. Protein A will bind the Fc portion of human IgG subclasses, IgM, IgA and IgE; and mouse IgG1 (weakly), IgG2a and IgG2b. Protein A also binds IgGs from other species, including monkey, rabbit, pig, guinea pig, dog and cat.
Immunoprecipitation Cells are lysed and cell debris are removed. A specific antibody is first added to the cell lysate. The Bio-Adembeads Protein A are then incubated with the cell lysate in order to form a magnetic labelled immune complex wich can be easily separated from the liquid by removing the supernatant during the magnetization step. Immunoglobulin purification Bio-Adembeads Protein A are particularly suitable for immunoglobulin purification from ascite, serum, tissue culture supernatants or other samples. After a short incubation time the immunoglobulins are eluted from the Bio-Adembeads Protein A and the containing Ig supernatant is pipetted off for downstream applications.
